Reporting Procedure

The Lactobacillus strains are reported as “Total Lactobacillus” and the Bifidobacterium strains as “Total Bifidobacterium.” These counts are reported in colony forming units (CFU)/gram on MRS agar (incubated at 37C for 72 hours). The “Total Lactobacillus” count is obtained by incubating the plates under aerobic conditions and counting all colonies; the “Total Bifidobacterium” count is obtained by incubating the plates anaerobically and then subtracting the Lactobacillus count from the total anaerobic count. This results in an approximation of the ratio of Lactobacilli to Bifidobacteria but is quite accurate with respect to the total probiotic count (Lactobacillus + Bifidobacteria).

Tests for Microbial Purity

Each production lot of Theralac® is sent to Minnesota Valley Testing Laboratories, Inc. in New Ulm, Minnesota for a microbial purity profile. The following tests are reported:

  • E. coli USP
  • Staphlococcus -- Coagulase Positive Confirmation
  • Total Mold Count
  • Total Yeast Count
  • Salmonella, BAM Method

Here are some tips for reading the purity analysis:

  • Each capsule of Theralac is approximately 450 mg or 1/2 gram.
  • The weight of the sample to test for E coli (for example) is 10 grams or 20 capsules.
  • A result showing negative means that in 20 capsules, no E. coli has been detected.
  • Other results may show <1 per gram (2 capsules).

CFU Plate Count Procedure

The procedures below demonstrate how we at Master Supplements, Inc. calculate the Colony Forming Units for TruFlora®.

  1. Aseptically empty 5 TruFlora® capsules into a sterile container, recording the weight from each capsule. Mix.
  2. Weigh out 1.0 gram of the mixed sample into a sterile stomacher bag.
  3. Aseptically add 99.0 mls of sterile, room temperature MRS broth (DIFCO) to the powder to make a 1:100 dilution.
  4. Stomach the sample for 5 minutes at high speed.
  5. Hold the sample for 30 minutes.
  6. Stomach the sample for 5 minutes at high speed.
  7. Serial dilutions are made in 0.1% peptone dilution blanks. Add 1.0 ml of the above dilution to 99.0 mls of diluent. Shake the dilution 100 times. This is the 10-4 dilution. Repeat the dilution and mixing step until the desired dilution factors are reached.
  8. For this product, plating the 10-8, 10-9, and 10-10 dilutions should cover the countable range.**
  9. Incubate plates at 37° C under aerobic conditions for 72 hours. **
  10. Count the colonies on each plate. Average the counts within a dilution. This gives the CFU/gram.
  11. Calculate the Colony Forming Units (CFU's) per capsule by multiplying the average count per dilution by the average weight per capsule from step #1.
